Skin toxicity during first-line enfortumab vedotin-pembrolizumab linked to treatment outcomes in single-center cohort
Retrospective · JAMA Netw Open · 2026-10-21. Number 5 in the UrologyWatch Top 5 for October 21, 2026.
Among 185 patients with advanced urothelial carcinoma receiving first-line enfortumab vedotin plus pembrolizumab, over half developed treatment-emergent skin toxicity (rash or pruritus), most commonly within two weeks of starting therapy.
Why it matters: Single-center retrospective data raise the question of whether early skin toxicity tracks with outcomes on enfortumab vedotin-pembrolizumab, warranting prospective validation.
Limitation: Single-center retrospective cohort; findings on the toxicity-outcome association may not generalize and causal direction cannot be established.
