PROTAC AR degrader bavdegalutamide shows activity in AR mutation subgroup in mCRPC

In this first-in-human phase I/II study of the PROTAC androgen receptor degrader bavdegalutamide (ARV-110) in mCRPC, patients with AR T878 and/or H875 mutations had PSA50 rate of 60% and median rPFS of 11.1 months, versus 4-5% PSA50 and shorter rPFS in other molecular subgroups.
Why it matters: Suggests AR mutation testing could identify a subset of heavily pretreated mCRPC patients most likely to benefit from this novel degrader class.
